In most places, you create a file in your home directory called .signature. Then you can either read the ASCII stuff into that file using your favorite editor (in emacs, use ctrl-X, i) and save the new file, or you can type the ASCII stuff directly in. Unless your service provider is weird, this will work. s/t/rn (and nn, I think) automagically append a file called ".signature" to the end of your posts, if it exists in your home directory. Elm, mail and mailx require that you edit your .mailrc (or elmrc) and specify whether or not you want a .sig included in your stuff. Elm lets you choose a local .sig and a remote one. If you need more basic information than what's been provided above, your service provider should have documentation or personnel that can help you.
